The Pentagon confirmed that B-52s took off from Andersen Air Force Base on the island.
"We cannot accept the US conducting nuclear strike drills targeting us and openly calling them strong warning messages," a spokesman for the North Korean military said on KCNA.
"The US should not forget that Andersen base... as well as other US naval bases in the Japanese mainland and Okinawa, are all within our precise firing range," the spokesman continued.
"If the enemy threatens us with nuclear weapons, we will respond with even more powerful nuclear strikes," North Korea added.
Previously, North Korea threatened a "preemptive" nuclear attack on the United States.
In a series of military activities that have escalated tensions on the Korean peninsula, this morning, the South Korean Unification Ministry said that North Korea has issued an airstrike alert, ordered its military to be ready for combat and asked people to seek shelter.
A spokesman for the Unification Ministry in Seoul said the alert, broadcast on national radio, could be part of a military exercise that North Korea has been conducting in recent weeks.
